Genomics Sequencing Policies


Summary

All samples to be sequenced should be submitted with a set of quality matrics to include an agarose gel (with sizing markers) and spectrophotography data.  Each sample requires its own library prep charge, but multiple samples can be mixed together per run. Each run is a separate charge depending on the sequencing requested.  Price includes labor, fluorometric determinations of quantity, and final library QC by BioAnalyzer and qPCR as needed.  Most paired-end sample libraries will be created with ~300 bp insert size.  Multiplexed samples may be mixed with other compatible samples.  Illumina now uses dual indexing libraries for all kits which are more expensive but provide more accurate de-multiplexing.

 

Sequence Data

All sequence data are live-streamed to the OSU HPCC "TIGER" cloud. Once sequencing is completed the reads will be demultiplexed, quality filtered, adapter trimmed, and converted into fastq format. Information on accessing your data by FTP or HTTPS will be emailed to you as soon as it is available. The TIGER cloud is only a temporary storage site, and all data is subject to deletion within about  4 weeks. For data stwardship, all sequence data are redundantly archived at the Oklahoma PetaStore at Oklahoma University, with a separate copy kept in Stillwater.

Samples

Sample amounts generally needed:

DNA: 100 ng to 1 ug in <30-40ul of 10mM Tris
RNA: 500 ng to 1 ug in <30-40ul of water

Smaller amounts of sample CAN BE SEQUENCED. Please contact us for experimental details.

 

We expect 400 million reads per run (high-throughput) and 130 million reads per run (mid-throughput) on OSU Genomics center-prepared libraries. The NextSeq500 has only ONE "lane" and therefore ONE "flowcell".

 

**NOTE** WE GUARANTEE 90% OF THE NUMBER OF READS SPECIFIED FOR THE INSTRUMENT.

We usually exceed the instrument's specified number of reads using Genomics center prepared libraries.

 

We will not provide "per read" pricing under any circumstances.

The number of reads is not guaranteed because variable sample quality cannot be fully controlled.  We will provide reasonable compensation for any errors mady by center personnel or malfunctions of the instrument itself.

 

We CANNOT guarantee a minimum yield of reads or base-pairs from a lane with customer-prepared libraries.

 

We perform quality control metrics on all samples which is not free. If your sample is unfit for library preparation or sequencing, you will be informed, and you will be charged for any additional QC.

If you request we run your sample regardless of quality, your samples cannot be mixed with other samples, so you will need to purchase a full run.

 

Pricing

The "Internal rate" is charged when payment comes from an Oklahoma State University account. All other service is charged at the external rate or commercial rate. Contact us with questions.

 

We pride ourselves in short turnaround times compared to commercial operations, however turn-around times will vary, and may be several weeks.

If you have more than seven samples they may be prepared robotically in batches of at least 8. One to seven samples can be done by hand at no additional cost.

 

If the sequencing instrument is unable to run, anyone with samples in the queue will be notified.
